OBJECTIVES
The objective of this work package is to assess the biodiversity of tropical pelagic species by using two molecular methods: eDNA (environmental DNA) and stomach content metabarcoding.
METHODOLOGIES
The methodology consists of two main parts: sampling and molecular analysis. We will first develop an optimal sampling protocol for both eDNA (seawater) and stomach content, testing different depths, distances from AFADs, and collection methods. For the molecular analysis, a specific gene (12S rRNA) will be amplified from the DNA samples, and the resulting sequences will be analyzed using bioinformatics to identify species. To improve accuracy, we will also create a local genetic reference database of fish species.
expected output
The expected output of this project is a comprehensive dataset of pelagic species detected through eDNA and stomach content metabarcoding. The project will also produce a finalized, optimal sampling protocol and contribute to the development of a local genetic reference database for fish.
